Das Thema seiner Dissertation lautete „Griechenland im Stadium der Industrialisierung“. Anschließend führte er seine Studien in Paris fort und wurde 1928 Dozent für Volkswirtschaftslehre an der Universität Athen. Im selben Jahr, im Alter von nur 24 Jahren, wurde er zum Professor für Volkswirtschaftslehre an der damals neu gegründeten Aristoteles-Universität Thessaloniki, wo er drei Jahre lang blieb. Er setzte dann seine Arbeit als Hochschulprofessor an der Universität Athen bis 1968 fort. In den späten 1940er Jahren übernahm er zusammen mit Ilias Lalaounis das renommierte Juweliergeschäft Zolotas aus Familienbesitz. Aufgrund politischer und akademischer Verpflichtungen verließ Xenophon Zolotas das Geschäft wieder.
Er war Vorstandsmitglied der UNRRA und hatte hohe Ämter im Internationalen Währungsfonds inne.
Zolotas war zwischen 1944 und 1945, 1955 und 1967 sowie 1974 und 1981 Direktor der Bank von Griechenland. Er veröffentlichte auch zahlreiche Arbeiten zu griechischen und internationalen Wirtschaftsthemen.
1952 wurde er in die Akademie von Athen gewählt. 1955 erwarb er auch einen Hochschulabschluss in Jura. Anschließend setzte er sein Studium an der Harvard University in den Vereinigten Staaten fort, wo er einen weiteren Doktortitel in Wirtschaftswissenschaften bekam. 1957 und 1959 hielt er in Washington zwei Reden, die als historisch gelten. Die Sprache dieser Reden war Englisch, jedoch verwendete er vorwiegend Wörter griechischen Ursprungs.<ref name=":0" />
1968 wurde er während der Diktatur von seiner Stelle als ordentlicher Professor für Volkswirtschaftslehre an der Universität Athen entfernt. 1974 übernahm er das Amt des Ministers für Koordinierung in der Übergangsregierung von Konstantinos Karamanlis, das er 1952 kurzzeitig bereits unter Dimitrios Kiousopoulos bekleidet hatte. Von 1984 bis 1988 trieb er als Finanzminister wichtige Wirtschaftsreformen voran, um die Inflation einzudämmen und das Haushaltsdefizit zu verringern.<ref name=":0" />
Als im Jahre 1989 weder die PASOK noch die Nea Dimokratia eine Mehrheit erreichen konnte, erklärte sich der damals 85-jährige bereit, bis zu Neuwahlen das Amt des Ministerpräsidenten auszuüben. Als bei den Wahlen 1990 die Nea Dimokratia unter Konstantinos Mitsotakis die Mehrheit erreichte, trat er zurück.

Berühmte Reden
Zwei seiner Reden im Englischen, die er als Vorsitzender der Bank von Griechenland am Abschlusstag der Internationalen Bank für Wiederaufbau und Entwicklung gab, erlangten große Aufmerksamkeit, da sie fast ausschließlich in Wörtern verfasst wurden, die aus dem Griechischen stammen.
